RET gene mutations cause MEN2, which is associated with medullary thyroid cancer, phaeochromocytoma, parathyroid hyperplasia and mucosal neuroma.
PTEN mutations cause Cowden syndrome, which does cause breast cancer (as well as thyroid cancer, endometrial cancer, macrocephaly and mucocutaneous lesions).
